Genetic structure of rainbow trout Oncorhynchus mykiss (Salmoniformes, Salmonidae) from aquaculture by DNA-markers

The rational use of valuable fish species from aquaculture is difficult to implement without knowledge the state genetic structure local stocks. Different types DNA markers can be used achieve goals selection and breeding work. a stock rainbow trout (Oncorhynchus mykiss Walbaum, 1792) (Salmoniformes, Salmonidae) farmed in Ukraine was studied using DNA-markers: microsatellite (SSR-markers – simple-sequence repeats-markers) intermicrosatellite (ISSR inter-simple sequence repeat). Five fragments trinucleotide motifs with single anchor nucleotide at 3'-end were as primer for analysis by ISSR-PCR method. Totally, 85 amplicons obtained across five loci, which 92.9% polymorphic. total number alleles ranged 10 (marker (ACC)?G) 23 (AGC)?G). following monomorphic determined trout: according marker (CTC)?C 770 520 bp bands, (GAG)?C 345, 295 260 bp, (AGC)?C 350 bp. average polymorphic bands per locus 15.8. selected ISSR primers had level information content above average. most effective molecular-genetic (AGC)?G percentage index, multiplex ratio resolving power. loci allow characterized (Na Ne 1.9 1.4, respectively), Shannon index (average value I 0.4) unbiased expected heterozygosity (mean uHe = 0.3). Microsatellite-based showed features six (OMM 1032, OMM 1077, 1088, Str 15, 60, 73). Allelic diversity established highest frequency typical given identified. SSR-markers 1.42 0.79, respectively. This depicts complexity population structure, high indicates stock. “gene pool profile” result future will help differentiate stocks Ukraine. Microsatellite provide ability determine individual variation populations conduct management resources on farms.
